K35 is a fluorescent probe for determination of albumin binding sites in plasma and serum [19, 20].

Square dyes exhibit high extinction coefficients and lower quantum yields in aqueous buffer but high quantum yields and long fluorescence lifetimes in presence of serum albumin. As an example Square-655 is a fluorescent probe with an extinction coefficient of 177 000 M-1cm-1 and a quantum yield of 80% in presence of albumin. These features make them the most sensitive fluorescent probes for the measurement of not only albumin but also other proteins. Because the binding constants of Square dyes to albumin are of the same order as for some of the common drugs these dyes are also useful for the estimation of drug-binding constants using the dye-displacement method.
