The Seta dyes series spans the whole visible range and the NIR.
SeTau-405-NHS (K7-547) is a dye that is well suited for excitation with the blue 404 diode laser. Its extinction coefficient is small but because of its large Stokes’ shift of more than 110 nm the quenching rates upon labeling to proteins are relatively low, which allows to obtain proteins with relatively high degree of labeling. Importantly the fluorescence lifetime of this dye is in the order of 9 ns, making it also a valuable probe for studying protein hydrodynamics (fluorescence polarization measurements). It is a good alternative for Pacific Orange to be used with Pacific Blue or V450 in flow cytometry applications. Biomolecules (antibodies, nucleic acids) labeled with this dye are well compatible with a Fluorescein emission filter. Exc./Em. max: 405/518 nm.
Our newly introduced fluorescent labels Seta-550, Seta-650 and Seta-750 are direct replacements of the conventional Cy series Cy3, Cy5 and Cy7 with more or less the same excitation and emission wavelengths as these but with higher quantum yields and photostability. Additional labels in this series will be released soon.
Seta-555-mono-NHS (K8-3335) is a direct replacement for Cy3 or Alexa 555 and can therefore be used with these filter sets. Its extinction coefficient is 154,000 M–1cm–1and the Q.Y. of Seta-555 is about twice as high as that of Cy3. Seta-580-mono-NHS (K5-3222) is a direct replacement of Alexa 580 or Cy3.5. It has a quantum yield of 55% and extiction coefficient of 90,000 M–1cm–1. It is well soluble in water and over all neutral in charge. Exc./Em. max for conjugates:580/605nm.
Seta-632-mono-NHS (K8-1642) is a better alternative to Alexa 633: firstly its absorption maximum is at 632 nm while that of Alexa 633 is blue-shifted to about 621 nm. Secondly Seta-632 has an extinction coefficient of 280,000 M–1cm–1as compared to 159,000 M–1cm–1 for Alexa 633. Emitting bright red fluorescence this dye can be measured with the Alexa 633 filter set. Exc./Em. max of IgG-conjugates: 637/647 nm. Seta-633-mono-NHS (K8-1663) is a hydrophilic, mono-reactive label for proteins and other biomolecules. It is extremely bright and has a low tendency for self-quenching even at higher D/P ratios. The extinction coefficient is 250,000 M–1cm–1as compared to 159,000 M–1cm–1 for Alexa 633, and quantum yields as high as 70% were measured with conjugates of this long-wavelength dye. It is optimally suited to be excited with the 633 nm spectral line of the He-Ne laser and the 635 nm line of red diode lasers and can be used instead of Alexa 633. Exc./Em. max of IgG-conjugates: 637/647 nm. Seta-635-NH-mono-NHS (K8-1669) is optimal for labeling of small molecules and oligonucleotides: its extinction coefficient is ~140,000 M–1cm–1, and the quantum yield of the free dye is in the order 0.31. It has a fluorescence lifetime of 1.8 ns in water (almost twice as long as Cy5) which makes it a very suitable dye for fluorescence polarization applications. It emits intense fluorescence in the far-red region of the light spectrum. Ex./Em. max: 639/655 nm. Seta-646-mono NHS (K8-1672) is extremely bright (extinction coefficient is ~210,000 M–1cm–1, and the quantum yield of labeled antibodies are as high as 33% at a D/P ~ 1) and an ideal replacement of Cy5TM, Alexa Fluor 647 or allophycocyanin producing intense red fluorescence emission. It is also extremely photostable and can be optimally excited with the 647 nm spectral line of the Krypton-Argon laser. Exc./Em. max for conjugates: 650/661 nm. Seta-660-di-NHS (K8-1682) is optimal if you are looking for a better alternative to Alexa 660: The extinction coefficient is 151,000 M–1cm–1as compared to 132,000 M–1cm–1 for Alexa 660, and quantum yields as high as 50% were measured from IgG-conjugates with this long-wavelength marker. It emits intense fluorescence in the far-red region of the light spectrum. Ex./Em. max: 661/672 nm. Seta-670-mono-NHS (K8-1342) is a dye that has been recently used in single molecule, homo-FRET measurements showed a remarkably low blinking effect which is an important factor in single molecule experiments. The dye has an extinction coefficient of ~180,000 M–1cm–1with an R0 for homo-FRET of 50 Å. Ex./Em. max: 667/686 nm. Seta-750-mono-NHS (K8-7035) is a hydrophilic, mono-reactive label with almost identical absorption and emission as Cy7 or Alexa 750 and can therefore be used with these filter sets. It combines high photostability and brightness. Its extinction coefficient is 220,000 M–1cm–1and the Q.Y. is about twice as high as that for Alexa 750. Ex./Em. max: 750/779 nm.
